The Samurai revolted against the Meiji Government because the government was taking away the values that the samurai upheld. As their values died, so did their honor. The samurai violently protested against these changes.

When the samurai rebelled the government sent the new imperial army to crush the rebellion.
It was very hard to defeat the samurai at first because the samurai did not fight in the open. As the next step, the government sent their navy but young, japanese men attacked the naval yard and seized all the supplies.

The samurai were defeated and the Meiji government did not have to worry about the any more samurai rebellions. The japanese army was fully modernized and many laws were changed because of all the fighting. Japan was no longer the secluded little island at the other end of the world.
Significance...
All the new reforms changed Japan. They no longer had their samurai and now depended on the newly trained imperial army to protect them. The changes also put an end to Japan's seclusion and Japan soon became a new center of trade.
